    孕酮是由黄体分泌的一种类固醇激素,食品中存在的孕酮对人体内分泌等健康产生严重影响。因此,探索简单、快速、灵敏的孕酮检测技术及方法很有必要。基于孕酮的特异性适配体(Apt)功能化的Au@Fe3O4纳米粒子构建磁弛豫传感器,在优化各种实验条件的基础上,对传感器的灵敏度、特异性进行评估,并对牛奶中孕酮检测的可行性进行分析。当孕酮适配体与Au@Fe3O4纳米粒子的体积比为3且孵育1.5h时,有利于Apt-Au@Fe3O4纳米探针的构建。磁弛豫检测过程中的优化条件如下:NaCl浓度为0.02mol/L,Apt-Au@Fe3O4纳米粒子稀释400倍,反应15min。在此条件下,水体系中单组份横向弛豫时间的变化量与孕酮浓度间呈良好的线性关系,线性区间为1~40nmol/L,检测限为4.97nmol/L,且具有良好的特异性。该方法可实现对牛奶样品中孕酮的检测,对牛奶中孕酮的线性检测区间为20~100nmol/L,检测限为13.60nmol/L,加标回收率为99.21%~103.45%,相对标准偏差不大于9.02%。

    Abstract:

    With the increase of public attention on food safety, it is important to establish a sensitive and rapid detection method for progesterone (pregn-4-ene-3,20-dione, P4) in animal food products. A magnetic relaxation switch (MRS) sensor based on aptamer functionalized Au@Fe3O4 nanoparticles (NPs) was developed for the detection of progesterone in milk. The test conditions were optimized to improve the sensitivity and stability of the method, and the feasibility of this method for the detection of progesterone in milk was also analyzed.The ratio of aptamers (Apt) to Au@Fe3O4 NPs, the incubation time of aptamers modification, the concentration of NaCl, the dilute ratio of Apt-Au@Fe3O4 and reaction time were optimized to be 3, 1.5 h, 0.02 mol/L, 400 and 15min, respectively. Under the optimized conditions, the change of mono-component relaxation time (ΔT2w) showed a linear range from 1nmol/L to 40nmol/L and the limitation of detection (LOD) was 4.97 nmol/L. In addition, the proposed method also illustrated an excellent specificity. The method was also applied successfully for the detection of progesterone in milk samples. The MRS displayed linear ranges of detecting progesterone from 20 nmol/L to 100 nmol/L and LOD of 13.60 nmol/L in milk. The average recoveries of progesterone in milk were 99.21%~103.45%, with relative standard deviations of nomore than 9.02%. The results indicated that this sensor could be a potential tool for the rapid detection of progesterone in food.
