    摘要:

    将高电压脉冲电场(PEF)技术用于牛骨胶原蛋白的提取,通过单因素试验和Box-Behnken中心设计法得出提取牛骨胶原蛋白的最佳工艺参数为:底物质量浓度1.0g/(100mL)、胃蛋白酶添加量3%、pH值2.5、场强21.98kV/cm、脉冲数7,在此条件下,可溶性蛋白质量浓度高达16.21mg/mL,通过紫外光谱鉴定了所提取的样品为纯胶原蛋白而不含其他杂蛋白。

    Abstract:

    Bovine bone was used as the raw material to extract collagen by the high-intensity pulsed electric fields. The optimal processing conditions were acquired by the single-factor experiments and the Box-Behnken experiment. The most yield of dissoluble collagen 16.21mg/mL was achieved under the following conditions: substrate concentration 1.0g/(100mL), pepsin concentration 3%, pH value 2.5, electric field intensity 21.98kV/cm, electric pulse number 7. The extracted collagen was identified by UV spectra that it was the pure collagen protein and did not contain other miscellaneous proteins. 
